
p-Nitrophenyl ions, -OPhNO2-p (PNP below), but not glycosides thereof, give a strong absorption at 405 nm. PNP-glycosides are useful for qualitative and quantitative studies of and with glycosidases, e.g. fucosidases, galactosidases and N-acetyl-glucosaminidases.
PNP-glycosides have also been used extensively for preparative synthesis of oligosaccharides with glycosidases (K. G. I. Nilsson, Trends in Biotechnol., 6, 256 (1988).
PNP-glycosides are also useful for covalent binding to proteins or solid phases after reduction of the nitro-group with e.g. Pd/C in ethanol to an amino-group.
PNP-glycosides may also be used for temporary anomeric protection after reduction to the amino-phenyl glycoside, acetylation of the amino group and subsequent treatment with CAN (patent pending).
